Speaker notes: Looking at the PK
of a single protease inhibitor there is the problem that, at the end
of a dosing interval, the concentration of drug in plasma falls to
a level close to the area of potential replication.
The terminology used varies somewhat: |

Cmin
or Ctrough: The lowest concentration reached during the
dosing interval
- tend to
be used interchangeably and can usually be considered to mean
the same thing
- there are
occasional exceptions to this rule eg nelfinavir (NFV) where Cmin
is not always the same as Ctrough.
IC50
or EC50 or IC95: The concentration required
to inhibit 50% or 95% of the virus, respectively
- generated
in vitro
- most pharmacologists
use IC50 as the most reliable parameter, but IC90
and IC95 are also used.
